In vitro cytotoxicity assessment and mechanistic insight of EpiOral™ tissue model to oral nicotine products with extraction stability analysis

Cell models applicable to the oral route of use are important for analysing cytotoxicity of oral nicotine products. MatTek™ EpiOral tissues were utilised when testing these products in the MTT [3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide] assay. Products were extracted in complete artificial saliva (CAS) to mimic product use. Extractions were performed on 300mg of product per mL in CAS, extracting for 2 hours. Treatments were performed on fresh extractions, 1-month frozen samples and 3-month frozen samples. Extracts were measured for nicotine and tobacco specific nitrosamines (TSNA), and analysis of frozen extracts was performed for each experiment. Lactate dehydrogenase (LDH), 8-isoprostane and cytokines release was measured to gather a mechanistic insight.
