An article in the October 2002 edition of "Toxicology in Vitro" summarizing the results of a European-based study to measure the predictive ability of in vitro skin models in assessing the skin irritation of cosmetics, has concluded that MatTek's EpiDerm Skin Model is an excellent in vitro model for this purpose. Further, the article, "Predictive Ability of Reconstructed Human Epidermis Equivalents for the Assessment of Skin Irritation of Cosmetics", also concludes that MTT ET-50 tissue viability assay endpoints provide the best in vitro-to-in vivo correlation.
This study is part of the European project, "Testing and improvement of reconstructed skin kits in order to elaborate European standards" (SMT4-CT 97-2174). The original work involved 4 in vitro skin models (MatTek's EpiDerm, L'Oreal's EPISKIN™, an in-house Cosmital model, and the SkinEthic™ Epidermal Model). Based on initial results, 3 of these models (EpiDerm, EPISKIN, and the Cosmital model) were selected to participate in the definitive round of testing.
The results from this multi-laboratory study involving L'Oreal and Wella Cosmital identified the MTT ET-50 tissue viability assay as the most predictive endpoint for skin irritation ("A comparison between in vivo and in vitro parameters resulted in the best rank correlation for ET-50, .."). The coefficient of correlation for MatTek's EpiDerm was a study-best R=0.94.
This study also concluded that while other assay endpoints may contribute additional information, MTT ET-50 endpoints were the most information-rich. Coincidentally, the MTT ET-50 is also the most cost-effective assay. In essence, the study found that the use of elaborate, time-consuming, expensive multiple endpoint assays were not necessary when measuring the skin irritation potential of cosmetics.
