373. PHOTOPROTECTIVE EFFICACY OF ISOFLAVONE GENISTEIN IN HUMAN RECONSTITUTED SKIN.

Moore, J.O., Gao, D., Wang, Y., Lebwohl, M.G., Wei, H. Mount Sinai School of Medicine, Department of Dermatology, New York, N.Y. Presented at Society of Toxicology 44th Annual Meeting, New Orleans, LA, March 6-10, (2005). (Published paper: See TR-422) Carcinogenesis, 27, 8, 1627-1635 (2006).

Summary:
This study by researchers in the Mount Sinai School of Medicine Department of Dermatology demonstrated that MatTek’s EpiDermFT full-thickness in vitro human skin tissue equivalent is an excellent in vitro alternative for studying the chemopreventive efficacy of topically applied anti-oxidants relative to UVB radiation exposure.

Genistein, the soy plant derived isoflavone, has garnered significant noteworthy attention from the scientific and medical communities alike in recent years. The recent heightened interest in reference to this naturally occurring soy derivative can be attributed to the positive beneficial health effects that increased soy consumption has been proven to have on disease risk assessment.

Genistein is a potent antioxidant and specific inhibitor of tyrosine kinase. Increasing evidence supports the preventive and therapeutic effects of this isoflavone in prostate and breast cancer as well as cardiovascular disease and post-menopausal syndrome.

Researchers in the Mount Sinai School of Medicine Department of Dermatology previously reported on the inhibitory efficacy of genistein against UVB induced photocarcinogenesis in Skh-1mice. This includes its capacity for scavenging reactive oxygen species, inhibiting photodynamic DNA damage, and the down regulation of signal transduction cascade pathways involved in skin cancer.

In this study they evaluated the capacity of topically applied genistein to modify UVB induced DNA damage and protect cell proliferation expression profiles in human reconstituted skin (EpiDermFT, MatTek Corp.).
